The Wallabies playmaker came off the bench on his return to the Tigers, coming on in the 49th minute with a commanding 49-7 lead.
O'Connor wrapped up the victory in the 76th minute, crossing for a try as they took down the Chiefs.
Former Waratah and Wallaby Izaia Perese also came off the bench for the Tigers, with former teammate Julian Heaven suiting up for the Chiefs.
Meanwhile, Angus Bell started for Ulster as they took down Edinburgh 40-19 in a rescheduled match from earlier in the year.
Bell put in a solid 60 minute shift to help them to victory, sitting third on the ladder.
Samu Kerevi and Israel Folau continue to fire in defeat for Urayasu D-Rocks.
The pair both had 10 tackle busts, yet were helpless to stop Toyota Verblitz as they went down 59-19.
It was better luck for Sean McMahon, who came off the bench for Tokyo Sungoltiah against Toshiba Brave Lupus.
McMahon joined former Red and new Japanese capped lock Harry Hockings in victory as they took out the 60-21 win.
As for Bernard Foley, his nine points were not enough as Kubota Spears lost in the final minute to Saitama Wild Knights in a 32-30 defeat.
